Hi I would like to know when the game 25620 ask me if I would like to activate the house card ability how would I use it's ability? Do I click proceed first cause there is no dialog box asking me to remove martell's house card

The Game State panel should include a description of what action you need to take. I believe With Melisandre you simply select a House card of your opponent by clicking on it, so that has a yellow frame. Then you click on proceed to submit your choice (you will lose a power token automatically).
If you click proceed without selecting a house card, you might be asked (I'm not sure about this) whether you really do not want to use the ability.
